(2019)LCMV Lymphocytic choriomeningitis virus is a rodent-borne pathogen belongs to Arenaviridae family. Most of the studies have referred Mus musculus as the main reservoir of the LCMV. It has been detected in pet rodents, laboratory rodents, and wild mice. Humans be infected with LCMV through the ingestion or inhalation of sources contaminated with rodent feces, urine, or both. LCMV infection can be asymptomatic, present with mild symptoms, or it can cause aseptic meningoencephalitis (AME) and teratogenic effects in infants. However, clinical cases of LCMV infection have been rarely reported, and there is only fragmental knowledge on the presence and prevalence of LCMV infections around the world. Likewise, the genetic characteristics of the circulating LCMV strains and impact of LCMV on public health have remained poorly characterized. This study was performed in the Southern Iraq, due to the lack of comprehensive information about LCMV in this area. There were three main aims in this thesis. First, to assess the prevalence of LCMV among the healthy human population in the Nasiriyah region, southern Iraq. Second, to assess whether LCMV infections can be associated with neurological manifestations. Third, to characterize the genetic variation and evolutionary history of LCMV strains circulating in southern Iraq. Serum and CSF samples were collected from patients and healthy people in Nasiriyah governorate in the Southern Iraq. Serum samples were screened for LCMV using Immunofluorescence assay (IFA) to detect IgG and IgM antibodies. Real-time PCR was used to detect LCMV genome. In order to confirm the PCR positive samples, we sequenced these samples by Next-generation sequencing. The serological assay results showed 12.22% IgG prevalence of LCMV among healthy people and 7.36% IgG prevalence among patients with neurological symptoms. The IgM prevalence was 1.25% among the patients with acute infections. From symptomatic patients, we sequenced partial L-segments of two new LCMV strains. The phylogenetic tree constructed on the basis of all known LCMV strains suggested that these new LCMV strains from Iraq are genetically distant from the previously known LCMV strains and form a novel sub-cluster within LCMV species. This study is the first survey of LCMV in the Southern Iraq. LCMV appears to be a rather common infection in Iraq. I reported new strains of LCMV that are circulating in the study site and most likely is the causative agent of the central nervous system-associated clinical manifestations in these patients. For future work, I’m aiming the detection of other Arenaviruses spreading in the Southern Iraq.

(University of HelsinkiHelsingin yliopistoHelsingfors universitet, 2005)Giardia on suoliston parasiittina elävä alkueläin, jota tavataan ihmisillä ja useilla muilla nisäkkäillä sekä sammakkoeläimillä, matelijoilla ja linnuilla. Nisäkkäillä esiintyvää G. intestinalis –lajia on tavattu ihmisen lisäksi mm. koiralla, kissalla, märehtijöillä, hevosella, rotalla ja majavalla. Tartunta tapahtuu feko-oraalisesti ulostesaastuneen juomaveden tai ruoan välityksellä tai kosketustartuntana eläimestä/ihmisestä toiseen. Koirilla infektio on usein oireeton, mutta se voi myös aiheuttaa voimakkuudeltaan vaihtelevaa ripulia. Muita mahdollisia oireita ovat laihtuminen, kasvun hidastuminen ja heikentynyt ruokahalu. Aikaisemmissa tutkimuksissa giardian prevalenssiksi koirilla on saatu 4-50 %. Suomessa prevalenssitutkimuksia ei ole aikaisemmin tehty koirilla. Tässä tutkimuksessa pyrittiin selvittämään giardian esiintyvyyttä suomalaisilla koirilla immunofluoresenssimikroskooppitutkimuksen avulla. Ulostenäytteitä kerättiin 146 koiralta. Näistä alle 6 kk ikäisiä pentuja oli 26 kpl. 10 koirista oli Yliopistolliseen eläinsairaalaan tuotuja löytökoiria. Muut olivat yksityisten ihmisten lemmikkejä. Kultakin koiralta pyrittiin saamaan kaksi ulostenäytettä 5-7 vrk välein. Giardia-positiivisten koirien osuus tässä aineistossa oli 4,8 %. Alle 6 kk ikäisillä koirilla giardia-positiivisten koirien osuus oli 15,4 % ja yli 6 kk ikäisillä koirilla 2,5 %.

(University of HelsinkiHelsingin yliopistoHelsingfors universitet, 2010)Borna-virus on Mononegavirales-lahkon Bornaviridae-heimoon kuuluva RNA-virus, joka aiheuttaa Bornan tautia. Bornan tauti on etenevä, usein kuolemaan johtava aivotulehdus. Tautiin on perinteisesti sairastunut hevosia ja lampaita maantieteellisesti rajoittuneella endeemisellä alueella Keski-Euroopassa. Nykytiedon mukaan Borna-viruksen isäntäeläinkirjo käsittää muitakin eläinlajeja, kuten lintuja, tautitapauksia todetaan eri puolilta maailmaa, ja viruksen on esitetty olevan osallisena myös ihmisten psykiatrisissa sairauksissa eli mahdollisesti zoonoottinen. Suomessa Borna-virusspesifisiä vasta-aineita on löydetty ihmiseltä, hevoselta, kissalta, koiralta ja villijyrsijöiltä. Kokeellisesti Borna-viruksella on saatu infektoitua lukuisia koe-eläimiä erilaisista jyrsijöistä lintuihin ja kädellisiin. Borna-viruksen epidemiologiaa ei tunneta täysin. Avoimia kysymyksiä ovat viruksen maailmanlaajuinen levinneisyys, isäntälajikirjo, leviämistapa ja mahdollisen reservoaarin olemassaolo. Reservoaarin olemassaoloa epäillään, koska Bornan taudin ei ole todettu leviävän helposti eläinyksilöstä toiseen, ja viruskantojen perimät ryhmittyvät sekvenssianalyyseissä maantieteellisesti eivätkä eläinlajin tai eristysvuoden mukaan. Villejä pikkunisäkkäitä pidetään todennäköisimpinä reservoaarilajeina, mitä tukevat vastikään havaitut Borna-virusinfektiot sveitsiläisissä päästäisissä ja suomalaisissa metsä- ja lapinmyyrissä. Metsämyyrä on Suomen yleisimpiin kuuluva nisäkäs. Se kantaa ja levittää tauteja, myös viruszoonooseja. Myyristä löydetyt Borna-virusvasta-aineet herättivät kysymyksen metsämyyrän roolista mahdollisena Borna-viruksen reservoaarina. Tämän tutkimuksen tarkoituksena oli selvittää 1) saadaanko metsämyyriä infektoitua kokeellisesti, ja jos saadaan, 2) millä infektioannoksella, ja 3) aiheuttaako infektio oireita ja/tai havaittavia vasta-aineita. Kokeeseen otettiin 15 paritettua metsämyyrää, jotka saivat yhteensä 52 poikasta. Poikaset infektoitiin aivoihin vuorokauden sisällä syntymästä käyttäen Borna-viruskannan He/80 4. rottapasaasista tehtyjä viruslaimennoksia 102, 103 ja 104 ffu. Myyränpoikaset lopetettiin ja elimet preparoitiin kahden, neljän, kuuden tai kahdeksan viikon kuluttua infektiosta. Aivonäytteistä etsittiin Borna-viruksen N-proteiinin geeniä käänteistranskriptio-PCR-menetelmällä ja verinäytteistä Borna-virusspesifisiä IgG-vasta-aineita immunofluoresenssimenetelmällä. Infektoiduista myyränpoikasista 24 prosenttia (10/42) oireili poikkeavasti verrattuna kontrollipoikasiin (n=9). Lähes kaikkien (40/42) infektoitujen myyränpoikasten, muttei yhdenkään verrokkimyyrän, aivonäytteistä monistui Borna-viruksen N-geeniä. Ensimmäiset positiiviset vasta-ainetulokset saatiin neljä viikkoa infektion jälkeen (p.i.), jolloin virusmäärällä 102 ffu havaittiin vasta-aineita 2/3 ryhmän myyristä ja virusmäärällä 103 ffu infektoiduista 1/5 myyristä. Kuusi viikkoa p.i. kaikki 103 ffu virusmäärällä infektoidut myyrät olivat vasta-ainepositiivisia, kun taas suurimmalla virusmäärällä 104 ffu infektoiduista poikasista havaittiin vasta-aineita vain puolella. Kaiken kaikkiaan vain 39 prosentilla infektoiduista poikasista havaittiin vasta-aineita veressä. Tutkimustulokset osoittavat, että myyrät infektoituvat Borna-viruksella, pääasiassa ilman oireita. Vaikka tulokset ovat vasta alustavia, ne tukevat metsämyyrän mahdollista asemaa reservoaarieläinlajina, ja antavat suuntaa luonnonjyrsijöistä saatujen vasta-ainelöydösten tulkintaan.
