(2023)Tämän kirjallisuuskatsauksen tavoitteena on kuvata dystrofiinin rooli ja merkitys lihaksen normaalissa toiminnassa, koota yhteen nykytietämys Duchennen lihasdystrofian patofysiologiasta sekä käydä läpi sairauden tutkimuksissa yleisimmin käytettäviä eläinmalleja. Duchennen lihasdystrofia on vakava, etenevä, perinnöllinen lihasrappeumasairaus, joka johtaa lihasheikkouteen, liikkumisen vaikeutumiseen, pyörätuolista riippuvuuteen, avustettuun hengitykseen ja lopulta ennenaikaiseen kuolemaan. Sairaus on poikalapsien yleisin neuromuskulaarinen sairaus, johon sairastuu noin 1/5000 syntyvästä poikalapsesta. Sairaus johtuu X-kromosomissa sijaitsevan dystrofiinigeenin mutaatioista, jonka seurauksena ei synny toimivaa dystrofiiniproteiinia. Ihmisten lisäksi homologiset sairaudet esiintyvät koiralla, hiirellä sekä kissalla. Dystrofiini ilmenee luusto- ja sydänlihaskudoksissa sekä pieninä määrinä aivoissa ja muualla kehossa. Lihassyissä dystrofiini sijaitsee lihassolukalvon alla, jossa se yhdistää solun tukirangan lihassolukalvoon sekä sitä ympäröivään tyvikalvoon. Dystrofiini muodostaa yhdessä muiden proteiinien kanssa dystrofiini-glykoproteiinikompleksin, joka toimii useissa tehtävissä solujen signaloinnissa sekä lihassolukalvon eheyden ylläpidossa lihassupistusten aikana. Ilman dystrofiiniä lihaksista tulee hyvin alttiita vaurioille, jonka seurauksena on etenevä lihaskudoksen korvautuminen side- ja rasvakudoksella, toiminnan heikkeneminen sekä kardiomyopatia. Sairauteen ei ole vielä olemassa parantavaa hoitoa, ja hoitojen tarkoituksena onkin oireiden kontrollointi sekä elämänlaadun parantaminen. Sairauden synnyn ja uusien hoitomuotojen etsimiseen hyödynnetään erilaisia eläinmalleja, joista osalla esiintyy spontaaneja mutaatioita dystrofiinigeenissä ja osa on geneettisesti muokattuja. Yleisimmin käytetyt eläinmallit ovat mdx-hiirimalli (engl. X-linked muscular dystrophy mouse) sekä kultaisennoutajan lihasdystrofia -malli. Dystrofiinin puutoksen aiheuttavia mutaatioita on kultaisennoutajan lisäksi todettu useilla muilla koiraroduilla, ja usein sairauden löytyminen uusissa roduissa on seurausta eläinlääkärissä heränneestä epäilystä.

(2020)Spinal cord injury (SCI) in human patients is the most expensive clinical condition worldwide, restricting individuals’ ability to manage with daily-life activities independently. With very limited available treatment possibilities, the understanding and validating of regenerative mechanisms and treatment options in animal models is crucial for their translation to clinical practice. The majority of SCIs in human patients are contusive in the cervical level of the spinal cord. However, thoracic injury rodent model is more commonly studied, with only recent studies working with cervical contusion injury model. Chondroitin sulphate proteoglycans (CSPGs), and especially their CS chains, are thought to be the major inhibitory structures for neurite regeneration after SCI. However, current research has led to a new idea that the inhibitory effect of CS chains can be reversed to regeneration enhancing by heparin-binding growth-associated molecule (HB-GAM). This endogenously secreted molecule is highly up-regulated in the central nervous system (CNS) during postnatal development, but in the adult CNS the expression is down-regulated. This suggests that postnatal-level concentrations might be needed for inducing neurite regeneration in adult CNS. In this study, HB-GAM treatment was tested on both cervical hemicontusion and hemisection injury models. Here we show that repeated intrathecal injections of HB-GAM were sufficient to increase grey matter myelin optical density in mouse hemicontusion injury model, and partly induce functional recovery in hemisection model. Obtained anatomical evidence suggests that enhanced myelination is potentially involved in the repair mechanism of HB-GAM. The connection between HB-GAM treatment and functional recovery, and also other mechanisms of HB-GAM-induced regeneration need further exploration. In broader perspective, the results are promising for translation of a novel treatment approach to clinical use.

(2013)Parkinson's disease is a neurodegenerative disease which is characterized by progressive loss of dopaminergic neurons in the substantia nigra and formation of intracellular Lewy bodies. α-synuclein is an essential part of Lewy bodies. In addition, mutations in the α-synuclein gene have been found to cause rare familial forms of Parkinson's disease. Animal models of Parkinson's disease are created by neurotoxins, transgenic animals and viral vectors. Transgenic animal models and viral vector models seem to reflect the pathology of Parkinson's disease better than the traditional neurotoxin models. In the transgenic animal models, the transgene and the promoter used in the expression of the transgene guide the pathology and motor dysfunctions that the animal model exhibits. In the viral vector models, it is important to use a suitable animal strain and a correct viral serotype in order to express the transgene sufficiently enough in the laboratory animals. The aim of the study was to investigate the ability of adeno-associated viral vector (AAV1-vector) to transfect WT- or A53T-α-synuclein gene into the striatum or the substantia nigra, and the effects of their overexpression on motor functions and concentrations of striatal dopamine and its metabolites in mice. In addition, the effect of a prolyl endopeptidase (PREP) inhibitor on the overexpression of A53T-α-synuclein in the mouse nigrostriatal pathway was studied, as PREP has been found to stimulate the aggregation of α-synuclein and therefore perhaps to increase neurotoxicity of α-synuclein. There were no statistically significant differences between the groups in the motor function tests (locomotor activity, rotarod and balance beam walk). Green fluorescent protein immunostaining showed that the GFP gene was weakly transfected into the striatum by the AAV1-vector, and no overexpression was observed. There were only minor differences in the striatal concentrations of dopamine and its metabolites. Finally, PREP-activity measurements showed that PREP-inhibitor (KYP-2047) treatment had poorly reduced PREP-activity. In this study, the viral vectors did not induce the overexpression of α-synuclein, although previously AAV2- and AAV6-vectors have been efficient in mice and rats. High PREP-activities that were found in most of the samples probably resulted from failed installations of mini-pumps that delivered the PREP-inhibitor. While in this study the viral vectors were not a successful attempt in the creation of an animal model of Parkinson's disease, they are an important method to model Parkinson's disease in the future.
