    Overall warming of climate, causing drought and sometimes co-occurring with insect outbreaks poses a risk to tree health and has been linked to tree mortality in many areas. The drought of 2003 was particularly severe in many regions in Europe, including Finland. It exceeded many previous drought years in its severity and intensity. Also, in the summer of 2006, 25% of the permanent forest sample plots of Finland National Forestry Inventory showed symptoms of drought damages. The Finnish south-west archipelago is high-risk drought area, and dry summers becoming more frequent and severe may cause severe forest damages in the area. In this study we investigated forest damages in Kemiönsaari, South-West Finland, approximately 60° 11'N, 22° 33'E. The forest damages there were first discovered in 2009, by local forest society, and has been spreading in the area. The crowns of Scots Pines (Pinus sylvestris) in the area were partially or completely defoliated. The defoliation started from the top of the canopy and spread evenly downwards. The central research questions of this study were: 1. Has the past drought years affected the growth of Pinus sylvestris in Kemiönsaari? 2. How is the Pine bark bug, Aradus cinnamomeus, affecting the trees hydraulic conductivity? 3. How is the Crumenulopsis sororia affecting the trees hydraulic conductivity? The findings provide support for the first research question stating that there has been a notable reduction in growth of the trees in the study area. The superposed epoch analysis revealed that droughts occurred in the past 20 years, reduced the radial growth of Scots pines. However, the results from this were within the lower 95% bootstrapped confidence limit and are not statistically significant. The results indicated that the most limiting factor to radial growth of Pinus sylvestris was precipitation in June, and last year July. Water shortage during the growing season decreases photosynthetic production and causes physiological stress to tree and is known to promote outbreaks of many insect. This study showed that the hydraulic conductivity of Scots pines in this area is not decreased by the pathogen Crumenulopsis sororia or Aradus cinnamomeus, in contrary, there was a clear correlation with higher hydraulic conductivity in the top of the crown of Scots pine’s where Crumenulopsis sororia or Aradus cinnamomeus were present.

    Finland has set ambitious goals to reduce road transport emissions. One of these goals includes halving emissions from transport by 2030 compared to the 2005 levels. Another goal is the carbon-neutrality target of 2035, in which the transport sector plays an important role as well. Achieving these targets requires efficient instruments. A possible instrument which can be used to reduce transport emissions sufficiently is emission trading. The possibility of creating a national emission trading system for the transport sector to reach Finland’s targets regarding emissions is the focus of this thesis. Establishing an emission trading system would likely have effects on the fuel price. I present the theoretical background before modelling these effects. Understanding how the fuel market works and how emission trading would affect it is crucial before analysing the impacts by modelling. The role of the price elasticity of demand in determining the change in the fuel price is highlighted. The existing literature has studied the topic of emission trading for transport extensively and points out three important design elements: the inclusion of the transport sector, the allocation of permits, and the point of regulation. These elements are discussed using previous studies on the topic. Furthermore, many studies highlight the possible distributional effects an emission trading system could cause. Exploring the currently operational emission trading systems around the world gives real-life examples of the different design elements and how distributional effects could be corrected in practice. When modelling the effects of emission trading on the fuel price, the value for the price elasticity of demand can be estimated using meta-analysis. For the meta-analysis, I selected 28 previous studies. The weighted average for the short-term elasticity is found to be -0.1607. For the long-term elasticity, the value is found to be -0.448. Using Monte Carlo simulations and the values obtained from the meta-analysis, it is possible to calculate the average changes in the fuel price and in the emission permit price, and also illustrate the uncertainty arising from the fact that the actual price elasticity of demand is not known. For emission reductions of 0.6 megatonne, the total change in the fuel price by 2030 is calculated to be 0.34 euros per litre. This means that the emission permit price would be approximately 204 euros per tonne of CO2 in 2030. In addition, other possible instruments as well as other emission reduction target values are modelled in the analysis. Finally, I discuss the design elements in more detail. An upstream system with auctioning would be an efficient option for the design of an emission trading system. In addition, I discuss the interaction of emission trading with other instruments and the possible EU ETS for transport. Last, I present limitations and some topics for future research.

    Oats are a good source of protein and dietary fibre, especially ?-glucan. Due to the health benefits of ?-glucan, oats have gained popularity in snack food formulations. The literature review deals with oats and its components, removal of lipids and fractionation of oat products. The particular emphasis of the literature review was given to studies on the effects of different oat fractions and extrusion process variables on the properties of oat-containing extrudates. The aim of this study was to find out how different oat fractions and extrusion process variables (screw speed, water content and feed rate) affect the physical and chemical properties of the extrudates. The measured physical properties were expansion, hardness and water content. Extrusion trials were carried out by using defatted oat endosperm flour (EF) as the main ingredient. Whole grain oat flour (WF) was used as a reference. To improve the nutritional quality, defatted oat protein concentrate (PC) and defatted oat bran concentrate (OBC) were added to EF. The oat fractions were defatted by supercritical carbon dioxide (SC-CO2). Pregelatinised corn starch (CS) and waxy corn starch (WS) were added in some trials to increase the expansion of the extrudates. A co-rotating twin-screw extruder was used for the extrusion. Different process variables were: water content of the mass (16, 18 and 20%), screw speed (240, 370 and 500 rpm) and feed rate (68, 76 and 84 g/min). The temperature profile of the extruder barrel was held constant in all of the trials: 40, 70, 70, 100, 110, 130 and 130 °C (sections 1 6 and die). Screw speed had significant effect on the expansion and hardness. Expansion increased and hardness decreased with increasing screw speed. Water content of the mass affected all the response variables in WF extrudates and all but not hardness and torque in EF extrudates. Increased water content of mass decreased the expansion and hardness in WF extrudates. Feed rate did not have significant effect on the physical properties. When using EF, more expanded and less hard extrudates compared to the WF were obtained. Addition of PC or OBC (10%) decreased the expansion and increased the hardness. Mixing of EF with corn starch (CS or WS; 30%) gave less hard and more expanded extrudates compared to pure EF. The highest expansion was achieved by the addition of WS. Even addition of OBC (20%) in a presence of WS (30%) gave highly expanded and less hard extrudates with high ?-glucan content 7.4% (dry weight). Decreasing the particle size of OBC (by ultra-fine milling) or the molecular weight of ?-glucan (by enzymatic hydrolysis) did not affect the physical properties of the extrudates even though small decrease in hardness was observed in the trial with enzyme-hydrolysed OBC. The results showed that defatted oat fractions can successfully be used in extrusion when mixed with corn starch. Screw speed had the most profound effect on the physical properties of the oat-containing extrudates followed by the water content of mass.

    Obesity and insulin resistance (IR) are key factors lead to equine metabolic syndrome and laminitis. Diet may play an important role in eliciting obesity by affecting insulin dynamics. Insulin-pathway signaling and mTORC1 genes may contribute to incred IR. The first objective of this study was to find and validate internal control genes for quantitative PCR method for adipose tissues in Finnhorse mares. The second aim was to quantitate the expression of mTORC1 and insulin-pathway associated genes after pasture season in two different treatment groups of Finnhorse mares and compare gene expression differences between treatment groups. In addition, gene expression differences were compared between two different adipose tissues. Twenty-two mares were equally divided into eleven equal pairs, the two mares of each group were randomly grazed either on cultivated high-yielding pasture (CG) or on semi-natural grassland (NG) from the end of May to the beginning of September. Eight pairs of Finnhorse mares were selected for gene expression profiling. Subcutaneous adipose tissue (SAT) samples were collected from two groups of Finnhorse mares after pasture season. Gene expression of neck and tailhead SAT were determined with quantitative Real-Time PCR method (qPCR). The selected internal control genes were actin beta (ACTB), glucuronidase beta (GUSB) and mitochondrial ribosomal protein L39 (MRPL39). Candidate genes were mechanistic target of rapamycin (MTOR), sterol regulatory element binding transcription factor 1 (SREBF1), sterol regulatory element binding transcription factor 2 (SREBF2), TBC1 domain family member 7 (TBC1D7), leptin (LEP), glucose transporter type 4 (GLUT4), monocyte chemoattractant protein-1 (MCP-1), retinol binding protein 4 (RBP4), tuberous sclerosis 1 (TSC1), tuberous sclerosis 2 (TSC2). There were no distinct gene expression differences between NG and CG groups in both neck and tailhead SAT. However, RBP4 had significantly (P=0.035) higher and GLUT4 had a trend (P=0.064) to higher mRNA expression in CG group in neck SAT. TSC1 had a trend (P=0.071) of higher expression in CG group in tailhead SAT. Gene expression differences were observed between tailhead and neck SAT. SREBF1 and GLUT4 had significantly (P=0.007 and P=0.026, respectively) higher expression levels in tailhead SAT compared to neck SAT. RBP4 had a trend (P=0.066) to higher expression in neck SAT compared to tailhead SAT. Minor differences in gene expression between NG and CG groups indicate that pasture-associated fat depositionmaynotconsiderably affect expressionof insulin-pathway and mTORC1 genes associated to obesity and IR in studied subcutaneous adipose tissues. These results also provide additional evidence to our hypothesis that fattening resulting on unrestricted grazing on cultivated high-yielding pasture does not increase the risk of metabolic diseases in Finnhorse mares when they have normal body condition at the beginning of the grazing season.

    Andean lupin (Lupinus mutabilis Sweet) is a potential oilseed crop, with a very high protein content (40–45%) and 15–20% oil content. It is valued as an alternative protein source for both human and animal consumption, and like several other lupin species, has a potential role in phytoremediation. Previous experience in central and southern Europe has shown the crop to have low and unstable yields with high sensitivity to heat and drought during the grain–filling stage. The species may therefore be more suitable for cool–temperate climates, as it comes from similar altitudes and latitudes as potato. Therefore, an experiment was set out to quantify the responses of Andean lupin to heat stress, using both gradually and suddenly rising temperatures during grain filling. In preliminary tests 60 accessions from 4 germplasm banks were screened for time to flowering and daylength sensitivity. For this experiment, 3 accessions were chosen, 478435, 457972, 457977, with vegetative phase durations of 44, 53, 64 days from sowing to first flower, respectively. Forty two plants of each accession were sown and reduced to 30 on the basis of uniformity. Plants were grown in a glasshouse with 22 °C, 18 h days and 18 °C, 6 h nights until about 25 days after flowering. Ten plants of each accession were subjected to one of the following treatments: control (continuing in the same glasshouse conditions), sudden heat stress (transferred to a growth chamber and subjected to 38 °C from 11:00 to 15:00) or gradual heat stress (transferred to a growth chamber and subjected to temperature increases of 4 °C day temperature and 2 °C night temperature, with the final two days at 38 °C from 11:00 to 15:00). The plants were returned to the glasshouse and when mature, the seeds were harvested and pooled into 3 replicates per accession and treatment for quality analysis. Seed protein, oil, soluble sugar, ash and moisture content were determined. Data were calculated on the basis of percentage of overall seed mass and also on a milligrams per seed basis in order to reflect the seed physiology at grain–filling stage. Sudden heat stress had greater effects on seed composition than gradual heat stress. When compared with control, sudden heat stress resulted in more loss of every component than gradual heat stress, on a per–seed basis, in all 3 accessions and the responses of the accessions to the sudden stress were not statistically different. Under sudden heat stress, mean seed weight declined by 70%, protein content by 70%, oil content by 85%, ash content by 50%, and soluble sugar content by 75%. The accessions responded differently, however, to the gradual heat stress. Accession 478435 experienced significantly greater reduction in seed weight, protein and ash content than accessions 457972 and 457977. Oil content per seed and soluble sugar content per seed were also lower in 478435 than in the other two cultivars, but the difference was not significant. On the flour basis, sudden heat stress increased ash content and decreased oil content and soluble sugar content significantly in all accessions. Accession 478435 had highest value in ash content at significant level. Under gradual stress, protein and ash content were increased while oil mass and soluble sugar mass were decreased. 478435 had significantly higher protein mass and ash mass in flour with respectively 57% and 5.1%, 457977 had significantly higher soluble sugar content with 112 mg/g. The results showed that heat stress can have a significant effect on the quantity and quality of seed yield in Andean lupin. While all tested accessions were severely susceptible to sudden heat stress, gradual stress identified differences between accessions, with one being much more susceptible than the other two. The most susceptible accession was the earliest to flower. Gradual heat stress allows better resolution than sudden heat stress when screening germplasm for heat tolerance.

    Fear of falling (FOF) and falling are urgent public health and economical challenges. Approxi-mately half of the elderly people experience FOF regardless of whether they had experienced fall or not. Increasing level of FOF can lead to self-induced restriction in mobility, physical func-tioning and activities of daily living (ADLs). Avoidance of activities weakens elderly's perfor-mance, well-being and increases the risk of falling. One-third of people over 65 years fall. For elderly 80 % of injuries are falls. Falls results in fractures, hospitalization, FOF, depression, loss of independence, premature residential care admission and even death in older persons. Number of falls has quadrupled over the past 25 years. Incidence of falling will increase in the future, as world population of people aged 65 and older is projected to increase to 997 million. Exercise is an essential method to maintain mobility, reduce FOF and prevent incidence of falling among community-dwelling elderly. “Strength in Old Age Programme” is a health enhancing exercise program (2005-2015) coordinated by the Age Institute. The project promotes independent living and quality of life (QOL) for elderly with weakened functional ability. A questionnaire was mailed to programmes’ participants consisting of questions about demographic and socio-economic status, health and well-being, physical activity habits and self-reported health improvements after participating the exercise group in April 2015. The aims of this study were 3-fold: first to identify biological, socio-economic, health and well-being, and behavioral factors affecting FOF and falling; second, to study perceived health improvements effect on FOF and falling; and last to investigate the complex association between FOF and falling. The key achievement was that “Strength in Old Age Programme” improved physical activity of elderly people involved in the study and affected biological, socio-economic and behavioral risk factors. This resulted in improvement in physical, functional, psychological and social domains reducing perceived FOF. Particularly good results in FOF reduction were seen in the participants who had higher age, were living alone and had lower socio-economic status. The incidence of falling was reduced through balance improvement in the exercise group. Similar yet smaller changes were observed in more vulnerable participants having poorer health and well-being related status. It would be important to influence the attitudes of elderly and motivate them proactively increase their physical activity to maintain balance, mobility, muscle strength, ADLs, health and well-being.

    The Atlantic forest on the Eastern coast of Brazil is one of the world’s most endangered biotopes. Less than 12 % of the original forest remains due to agricultural and pasture expansion. In addition, many Atlantic forest restoration projects in the past have failed, largely because of inadequate silvicultural practices. Meanwhile, the growth rates of Eucalyptus and pine plantations have been increased 3 to 4 fold in Brazil over the last four decades by utilizing intensive silvicultural methods such as site preparation, fertilization, and weed control which in turn increases resource supply (nutrient, water and light). This study determines the effects of these same intensive silvicultural methods on Atlantic forest restoration regarding initial growth and carbon sequestering. Two parallel research sites were established in 2004 on latitudes 11°S and 23°S on the Eastern coast of Brazil to determine the effects of intensive silviculture, planting density and species composition on the development of 20 native tree species. This research focused on the Northern site (200 km North of Salvador, Bahia State) which has a typical tropical climate and soil type. The project has a 23 factorial design totalling 8 treatments, with the following factors: i) intensive and traditional treatments; ii) initial planting densities (3333 trees ha-1 and 1667 trees ha-1); and iii) species composition proportion (50:50 and 67:33 ratio of pioneer vs. late successional species). After 8 years from planting, survival and development of each species, aboveground biomass and leaf area index (LAI) were determined for all the treatments to compare the effects of the different factors. In summary, the main findings of this study are: 1) The more intensive management methods improved survival and the initial growth of tree species 2) Lower stand density (1667 trees ha-1) had the best response to the intensive management for LAI, stemwood production, and above ground carbon sequestration 3) Out of 20 species, 19 had significantly higher growth with intensive management, indicating that both pioneer and late successional species are constrained by the original site conditions. 4) Intensive management was essential, especially for non-pioneer species. 5) Under low intensity silviculture, the 67:33 ratio pioneers vs. non-pioneers with higher planting density (3333 trees ha-1) was the best option to obtain the highest stemwood volumes 8 years from planting, while the 50:50 ratio pioneers vs. non-pioneers with lower planting density (1667 tree ha-1) could be recommended under intensive silviculture. Conclusion: Intensive management methods have the potential to increase early restoration success by increasing biodiversity through enchancing survival and growth of non-pioneer species and accelerating the canopy closure. Intensive management methods increased the above ground carbon sequestered in 8 years, remarkably, up to 3-fold compared to traditional management, making it an attractive management option for carbon offsets.

    Herbs are valued for culinary and health purposes and their metabolism and chemical composition can be influenced with LED lighting. This Master’s Thesis aimed to study how different spectra (green, blue, and white light) affect the sensory properties of hydroponically grown dill (Anethum graveolens L.) and coriander (Coriandrum sativum). The hypothesis was that green light produces more soapy and musty flavours in coriander, whereas blue light produces more citrus and typical coriander-like flavours. For dill the hypothesis was that blue and green light treatments produce stronger flavours compared to white light. A generic descriptive analysis method was chosen, and trained panels created sensory profiles for three light treatment and one commercial coriander and dill samples. Intensities of smell, taste and flavour attributes were evaluated using a line scale (0 = not at all to 10 = extremely) in three replicates. Study was conducted during the COVID-19 pandemic in the sensory laboratory conditions (ISO 8589). One-way ANOVA showed that light treatments had only slight impact on the sensory profiles of coriander and dill. In coriander blue light produced significantly lower lemon odour intensity compared to green light treatment. In dill total odour intensity was significantly lower in blue light sample compared to white light and commercial samples. Otherwise, one-way ANOVA did not show significant differences between samples. However, principal component analysis (PCA) implied that samples differed. Two-way ANOVA results showed that neither panel worked uniformly and deviation among intensity scores was observed. Herb samples proved to be rather difficult to evaluate and more extensive training could have improved panel’s performance. In future consumer study could be performed to study if spectrum affects the hedonic response to these herbs.

    Applying organic amendments to agricultural soils holds a potential to mitigate climate change by increasing soil carbon stock. It may also provide other benefits, such as enhanced nutrient cycling along with improved soil quality and fertility. Aim of this study was to determine whether utilizing organic soil amendments has effects on soil aggregates and root-associated fungi, both of which are associated to increasing soil carbon stocks. Soil and oat (Avena sativa L.) samples were obtained from a field experiment established in Parainen, South-West Finland in 2016 on a clay (54 % clay) soil. Studied treatments included four organic soil amendments (two wood-based biochars and two fibrous by-products of paper industry), mineral fertilization control soil and untreated control soil. Sampling was done two years after the amendment application, in 2018. Soil sampling was done after the harvest and plant sampling at three timepoints (June, July and August). Soil aggregates were separated from the bulk soil by wet sieving to determine their stability and size distribution. Carbon content of aggregates, as well as topsoil (0-5 cm) carbon stocks were quantified. Oat roots were stained and colonisation rate of different intra-root structures formed by root-associated fungi (arbuscular mycorrhizal fungi and dark septate endophytes) determined with microscope. Studied organic soil amendments did not affect soil aggregate structure or carbon content of aggregates. Nevertheless, C/N-ratio of some of the studied samples as well as larger soil carbon stock imply that other one of the studied biochars could have increased soil carbon content. When observing fungal data by month for between treatment differences, AM arbuscules were found to be most common in untreated control soil in August. When monthly fungal structure observations from all the studied treatments were combined, clear change in intra-root structures was detected (likely due to seasonal dynamics of fungi); AM arbuscules were more common in June and July, whereas DSE structures predominated in August. Large variation in the field together with methodological constraints might hide some effects of organic amendments and to verify their impacts, more research is needed. Based on the results gained in this study, effect of organic soil amendments seemed to be neutral on both soil aggregates and root-associated fungi. Since no adverse effects were detected, studied soil organic amendments could be utilized as recycled fertilizers.

    Metabolic disorders are known to predispose dairy cows to periparturient diseases. Main components of metabolic disorder are insulin resistance and severe negative energy balance which are responsible for the reduced fertility in addition of increased risk of disease in dairy cows. The mobilization of adipose tissue in response to energy deficiency is associated with metabolic and endocrine changes during early lactation. Reduced insulin sensitivity in peripheral tissues could potentially change the relative rates of lipolysis and lipogenesis. This study focused on expression of lipogenesis and lipolysis associated genes around parturition. Samples and data sets for this study were obtained from the feeding experiments conducted from September 2010 to April 2011 in Viikki Experimental Farm at the University of Helsinki. Sixteen multiparous Ayrshire cows were divided into two groups based on energy level and fibre content of their feed: (1) grass silage group (control) and (2) silage-roughage mixture group (experiment). During experimental period average energy intake (MJ/day) in silage group was 35% higher than in silage-roughage mixture group. Subcutaneous adipose tissue samples were collected a week before, one day and a week after parturition from cows. Total RNA was extracted from tissue samples quality and quantity of total RNA was analysed using electrophoresis and spectrophotometer. Complementary DNA (cDNA) was prepared from the total RNA for quantitative PCR (qPCR). QPCR was conducted to quantitate expression of the following genes: adiponectin (ADIPOQ), leptin (LEP), peroksisome proliferator activated receptor gamma (PPAR-?), adiponectin receptor-1 (AR1), adiponectin receptor-2 (AR2), lipoprotein lipase (LPL), stearoyl-CoA desaturase (SCD) and hormone-sensitive lipase (HSL). Two genes, AR2 and LEP, were downregulated in group 1. Reduced expression of AR2 in group 1 may relate to an increased insulin resistance. The glucose metabolism was reduced further leading to reduced insulin sensitivity. Lower expression of LEP after parturition indicates usage of energy for milk production. The upregulation of SCD in group 1 before and after calving as well as after calving in group 2 may be a result of the uptake of fatty acids by the mammary tissues. The expression of ADIPOQ, AR1, LPL, PPAR, and HSL did not show any significant changes.

    The faba bean (Vicia faba L.) has increased its cultivation area in Finland because of its high protein content. It replaces some of the imported plant proteins, such as soybean (Glycine max) meal, as feed for livestock and has increasing interest as an ingredient in plant-based meat-substitute food products. The faba bean is partly self-pollinating, but insect pollination has been shown to increase yield and reliability. Honeybees (Apis mellifera) are one of the most important pollinators for the bean in Finland. The aims of the thesis were to study the effects of honeybee interactions with faba bean flowers and pollination. This was done mainly via a cage experiment about the pod count and distribution along the plant stem, with and without pollination from honeybees. The second and third aims were to find out more about the honeybee behaviour on the faba bean flowers and the abundance of pollinators and species richness on faba bean fields. For these parts a field survey was conducted on eight different faba bean fields in Southern Finland. The increase in pod count following bee activity was found to be as high as 65% and the increase in pods per node 19%. The distribution of the pods was increased towards the middle of the plant in the plants that were insect pollinated, compared to the plants that were not. There was a strong correlation between the number of honeybees (Apis mellifera) on the fields and the pod counts per plant node. Honeybees visited 1.5 flowers per plant on average and frontal visits constituted 55% of all visits recorded. Pollinator numbers and species diversity varied greatly from field to field. The results show that pollination by of honeybees benefits faba bean yield. They give a good understanding into the potential yield effects and provide pointers for future research on the topic. The study is aimed to prepare the background for introducing a better basis for cooperation between beekeepers and faba bean farmers. The main goal is to increase knowledge about honeybee pollination effects on the faba bean yield and give a solid start into building prospects for commercial pollination services in Finnish agriculture in general. The study was done in collaboration with The Finnish Beekeeping Association.

    Parkinson’s disease (PD) is a neurological disease impacting over six million people in the world. In addition to the motor symptoms of the disease, the disorder is accompanied with gastrointestinal problems. These include delayed transit time, disturbances in the microbial composition of the gut, inflammation in the intestine, structural changes in the epithelial lining and increased intestinal permeability. Paracellular permeability is regulated by tight junctions (TJ) which are complexes formed by proteins such as claudins, occludin and zonula occludens proteins. Probiotics are live microorganisms which when administered have beneficial effects on the host. Probiotic bacteria can modulate intestinal permeability and the expression of TJ proteins. These microbes are also able to improve the gastrointestinal symptoms, such as constipation, in PD patients. As a part of a larger study, the aim was to examine whether the expression of TJ proteins is altered in the lactacystin-induced mouse model of PD and to investigate whether probiotic supplementation elicits changes in their expression. The study was conducted in eight- to nine-week-old C57Bl/6JRccHsd mice, to which PD symptoms had been induced by lactacystin injection to the substantia nigra. Starting at one to two weeks after the injection, four groups of mice were treated with one out of four specific probiotic strains (A, B, C and D) for two weeks. At the end of the experiment, jejunal and colonic samples were collected from the mice. These samples were analyzed using Western Blot to determine the expression of TJ proteins claudin-1, claudin-4 and occludin. Based on the WB results, the jejunal gene level expression of these proteins in one probiotic group (D) was compared to the untreated group with reverse transcription quantitative polymerase chain reaction. The expression of TJ proteins was not altered as a result of the lactacystin injection in jejunum or colon, suggesting that lactacystin model is not optimal for investigating PD-related alterations in TJ proteins. Supplementation with microbe D increased the jejunal expression of claudin-1 on both protein and gene level, whereas in the colonic protein expression there was no change. Clear effects were not detected on claudin-4 or occludin nor when treated with microbes A, B or C. Therefore, microbe D might influence intestinal TJ function and permeability through regulating the expression of claudin-1.

    Gastrointestinal parasites (GIP) and resistance against anthelmintics are and will be one of the biggest challenges for the sheep husbandry. Research within this area is very important today and it is important to figure out new methods to identify individuals with high burden of parasites and to treat just these individuals. The aim of the study was to investigate the effects of GIP on activity patterns and weight gain in first season grazing lambs. A sub-study accompanied with the aim was to see if the manually made behavioural observations supported data from the sensors. The study took place at the Swedish University of Agricultural Sciences (SLU) during the summer of 2019. The study was divided into two experimental periods. In the first experimental period 30 ewes and their 60 twins were used. Of those, 58 lambs continued to the second experimental period. Two groups were dewormed with ivermectin (high group) and two groups were untreated (low group). All lambs were fitted with sensors, and the data was downloaded once a week, at the same time with the weighting of animals. Individual faecal samples were collected four times. The pasture where the lambs grazed was a first-year pasture. Sward height was measured, and herbage samples were collected three times during the experiment. Herbage samples were analysed for nutritional and mineral content. The manual behavioural observations were made two days a week during five weeks. The high group had higher weight (28.9 kg) in the beginning of the experiment, compared with the low group (28.1 kg). On the contrary, the high group had lower weight (42.7 kg), compared with the low group (44.8 kg) at the end of the experiment. According to data from the sensors there were differences in lying time and motion index (MI) during the 7-10 first days after weaning. The low group laid down more, compared with the high group. The low group had higher MI compared with the high group. During the whole experiment there were no differences in lying time, lying bouts or MI. The manual behavioural observations showed that “standing still” had significantly more registrations for the high group compared to the low group. It is possible to see differences in behaviour with sensors at an early stage of parasitic infection, even with low levels of parasites. This means, that there is a potential to use automatic behaviour observations as a diagnostics tool during infection of GIP.

    The feeding behavior of beef cattle is complex and varies depending on different factors such as the nutritive value of the feed, the individual and the environment. By optimizing and improving the feed efficiency, the economics of the production increases. Knowledge of feed intake and feeding behavior can be used to optimize the production and the health and well-being of the animals. The aim of this study was to determine how the cut of grass silage affects the feeding behaviour of bulls and thereby how to optimize the use of silage for animal growth. Simmental bulls got three different silage cuts. The first cut was taken on 25th June (ES1), the second cut on 11th August (ES2) and the third cut on 3rd October (ES3). The feeding was given as total mixed ration (TMR) ad libitum. Each TMR group had 15 bulls (TES1, TES2, TES3). Each TMR group had a diet based on grass silage (550 g/kg DM), the only difference being if the silage was of the first, second or third cut. That way the nutritive value differed among the TMR. Individual information about duration, when, and how much the bulls ate was recorded by the Growsafesystem. The average for the bulls in the feeding groups was calculated based on the individual data. This Master´s thesis researched feed intake, feeding frequency, time for head down, time for feed intake and feed intake rate during one month when the body weight of the bulls was on average 546 kg in the beginning and 607 kg at the end of the study. The dry matter for the three cuts was 201 g/kg for ES1, 298 g/kg for ES2 and 354 g/kg for ES3. D-value was 719, 685 and 739 g/kg DM, respectively. The bulls that ate ES3 had higher dry matter intake (12,1 kg DM/d) and eating rate (94,9 g DM/min) than the two other groups. The bulls that ate ES1 and ES2 had the same feed utilization, which was better than the feed utuilization of the bulls that were given ES3. Feeding frequency was about 15 times per day for all groups. The second cut had the lowest digestibility and the bulls eating the second cut had the longest feed intake time. The bulls that ate the third cut had the highest dry matter intake, but the lowest feed utilization. The study indicates that the D-value of the grass silage used in TMR has a clear impact on dry matter intake of the TMR and eating time and eating rate of the animal.

    Garden strawberry (Fragaria x ananassa Duch.) is one of the most important soft fruit crops in Finland. Every year, over ten thousand tonnes of strawberries are produced, but in many cases they are affected by grey mould disease (Botrytis cinerea Pers.). Grey mould disease is absolutely devastating for strawberry, as it can affect up to 80% of the yield. Although chemical fungicides are widely used in controlling the pathogen, there are increasing concerns about its safety and environmental impacts. The use of disease resistant cultivars and biological control agents (BCAs) are two of the alternative methods in controlling grey mould disease. However, the breeding progress of resistant cultivars is limited by the selection methods used, while the BCAs require more characterization. In this study, we evaluated strawberry grey mould resistance by inoculation, and tested the amount of Pseudomonas fluorescens in strawberry after biological control application. Inoculation of strawberry is feasible, efficient and reliable to replace the conventional grey mould evaluation method. Foliage application of P. fluorescens did not significantly change the endophyte content of strawberry, proposing that the use of the bacterium is safe from causing any bacterial contamination.

    Tutkielma käsittelee tulevaisuuden ruokaa ja rakentaa käsitystä siitä, miten kuluttajat suhtautuvat tutkimuksessa käsiteltyihin uusiin raaka-aineisiin, ruokiin sekä ruokateknologiseen kehitykseen. Tutkielmassa analysoidaan kuluttajien suhtautumista geenimuunnettuun ruokaan, hyönteisten syöntiin ja mahdolliseen keinolihatuotantoon. Tämän lisäksi tarkastellaan sitä, minkälaisia pohdintoja ja mielikuvia haastateltavat liittävät tulevaisuuden ruokaan. Aihetta lähestytään sosiologisen ruokatutkimuksen perinteen kautta. Tutkimusaineisto koostuu 20-35 vuotiaille suomalaisille korkeakouluopiskelijoille tehdyistä viidestä teemahaastattelusta, jotka toteutettiin pari- ja pienryhmähaastatteluina. Haastatteluaineisto on analysoitu käyttämällä teemoittelua. Tutkimuksen mukaan kuluttajat suhtautuvat geenimuunnettuun ruokaan epäilevästi ja liittävät siihen terveyden, ruokateknologian ja epätietoisuuden teemoja. Pelko negatiivisista pitkäaikaisista terveysvaikutuksista on korostunut geenitekniikkaan liitettyjen epäilyjen lisäksi. Hyönteisten syöntiin suhtaudutaan myönteisesti ja hyönteisten uskotaan olevan tulevaisuudessa käytetty raaka-aine ja potentiaalinen lihankorvike. Kuluttajat eivät halua nähdä tai syödä hyönteisiä kokonaisina, mutta jauheena tai muun ruoan osana hyönteiset ovat hyväksyttäviä. Keinolihaa pidetään turhana innovaationa ja siihen liitetään epäilyjä ruokaturvallisuuden ja ruoan puhtauden osalta. Kuluttajat liittävät tulevaisuuden ruokaan pohdintoja luonnonmukaisuudesta ja keinotekoisuudesta sekä niihin liittyvistä vastakkainasetteluista. Myös liha- ja kasvispainotteisen ruokavalion vertailu sekä luottamuksen merkitys koetaan tärkeänä. Geenimuunnettuja ruokia ja keinolihaa pidetään keinotekoisina ruokina, kun taas hyönteiset nähdään selkeästi luonnonmukaisimpana vaihtoehtona. Lihan kulutuksen uskotaan vähenevän tulevaisuudessa ja kasvispainotteisten ruokavalioiden lisääntyvän. Luottamus nähdään tärkeänä tekijänä ruokasektorin jokaisella osa-alueella ja varsinkin uusien ruokien ja teknologioiden tapauksissa. Tutkimuksessa esitetään, että kuluttajat kokevat geenimuunnetun ruoan ja keinolihan teknologisina ja keinotekoisina tuotteina, joiden hyväksyttävyys osaksi omaa ruokavaliota tai yhteiskuntaa voidaan kyseenalaistaa. Hyönteiset taas nähdään luonnonmukaisena ja periaatteessa hyväksyttävänä raaka-aineena ja ruokana. Tutkimus nostaa esiin ruoan kesyyntymisen tärkeyden uusien raaka-aineiden ja ruokien kohdalla. Kyse on prosessiluonteisesta uusien ruokien kesyttämisestä, jotka on neuvoteltava osaksi omaa elämää ja yhteiskunnallista arvostusta. Kuluttajien tulee myös luottaa elintarvikealan toimijoihin, jotta uudet tuotteet voidaan hyväksyä markkinoille.

    ABSTRACT Background and aim of the study: Recently the reliability and accuracy of traditional research methods of food intake have been questioned. Traditional methods have a tendency to underestimate the energy intake of the subject. Individuals’ ability to estimate the portion size, memory, and self-report affects the reliability. New technology offers new tools for nutritional research. Individuals’ food and energy intake can in theory be estimated from photographs showing the food portions and left over food. In this master’s thesis the aim is to investigate if food photographs of preschool children can be used in assessment of food consumption. The reference method in this research was weighted portion sizes. Active food photography method has not in fact been investigated on small children before. Design and methods: This master’s thesis is part of the DAGIS research project. The subjects (n=22) were children aged 3–6 years in two day care centers in Helsinki. The food intake of the subjects was photographed during lunch hour and snack time in the daycare setting with two cameras. All food items and left over food was also weighed. Research material includes also the children’s food diaries filled in by the daycare personnel. Portion size estimates from food photographs and food diaries were compared against the weighted portion sizes. Arithmetic means and standard deviations were calculated for different food groups. The difference between the weighed portion size and the estimated portion size was calculated with 95 % confidence interval. The geometric means with 95 % limits of agreement were also calculated for the ratio of weighted and estimated portion size. Bland-Altman figures were drawn to assess the agreement between methods. Results: This master’s thesis found that on average food photographs overestimated the portion sizes compared to the weighted portions (geometric mean 1,06). The estimations made from the photographs were in line with the food records in if they over or under estimated the portions in different food groups. When comparing the energy intake from the study meals this study found that there was no difference between the estimated energy intake from photographs and the energy intake from weighted portions (geometric mean 1,01). The differences made in the portion size estimation (over or underestimation) were seen only in part of the food groups in both estimation methods. Conclusion: This study found no difference between the energy intake estimated from the food photographs and weighed portions. This was an exceptionally good result compared to earlier studies where food photographs have underestimated subjects’ energy intake. The explanation for the difference in results may possibly be explained by the study design. In this study the food photographs were taken at the day care centers by researchers and the day care personnel filled in the food records. Third party report is different from self-report. Food photographs have hardly been studied in food consumption studies with small children before. The heterogeneity of the previous studies and the lack of validation of the different food photograph methods make the comparison of the study results difficult. This study suggests that food photographs could someday even replace the traditional methods. Before that can happen, we need more studies where the method has been validated and the study settings is well planned forehand. Food photographs would be a useful tool for nutritional studies especially in a busy day care environment.     Agriculture and other human activity has considerably affected the nitrogen cycle, which initially doubled the amount of nitrogen on the planet. The most well-known consequence of this phenomenon is eutrophication. The objective of this thesis was to study nutrient balances and their usefulness in evaluating the nitrogen usage in beef cattle farms, the nitrogen load and nitrogen use efficiency in those farms as well as how well the farms utilize their inputs. This study based on three organic beef cattle farms which took part in BERAS Implementation –project in 2010-2013. All farms specialized in suckler cow production. The data was collected in July-August 2013 when the farmers were interviewed about their inputs and outputs during the years 2010-2012. On the basis of this information different type of nitrogen balances were calculated. The farm gate nitrogen balances were 27, 36 and 70 kg/ha. Except for the last one, the feed self-sufficiency of the farms was very high, more than 90 %. The farm with the largest nitrogen surplus utilized a considerable amount of input from outside the farm which reflected in low feed self-sufficiency (64 %). The other two with smaller surpluses received excellent primary nutrient ratios (1,38 and 1,40) which indicates active nutrient recycling. However, despite of these results, it is important to remember that nutrient balances should be viewed with a critical eye. The uncertainties primarily concern the amount of biologically fixated nitrogen and the amount of yields of cultivated plants. These variables were taken into consideration in sensitivity analysis. Instead of nutrient balances, it would more useful to draw attention to other indicators such as feed self-sufficiency or animal unit density when evaluating nitrogen load of farms. The efficiency of beef cattle production depends also on how the production is actually implemented. It is especially important to acknowledge the importance of feeding and manure handling. The feed conversion ratio is always low in beef production since it is less than 10 %. Nevertheless, there are other benefits in cattle breeding such as utilization of natural pastures and cultivation of perennial grasses which not only are excellent for soil fertility, but also decrease nutrient leaching.

    Wetlands are one of the most diverse ecosystems in the world. They provide numerous different ecosystem services and their importance has been acknowledged internationally. However, the number and extent of wetlands has continued decreasing globally and the increasing population poses threats on more and more wetlands. The Life+ Urban Oases project (fin. Keidas-hanke), that has made this Masters Thesis possible, has pursued to bring wetlands into urban areas, where the ecosystem services, such as flood control, which they provide can be utilized to compensate for challenges brought about by changes in land use. Urban wetlands provide local population recreational and relaxing green oases. Urban wetlands also provide natural system stormwater management mitigating water quality and habitats. It is predicted that climate change will increase severity of weather extremes. Especially the probability of heavy rainstorms is expected to increase in Finland which will emphasize better stormwater management. The value of ecosystem services provided by stormwater wetlands has not been previously measured in Finland. Furthermore, beliefs of people on constructed or created wetlands in general has been studied very little. This study had two objectives: 1) To evaluate the value of the ecosystem services of constructed urban and agricultural wetlands within the Municipality of Vihti. This was carried out within the Lake Enäjärvi watershed. 2) To study what are the attitudes, perceptions and beliefs of the Vihti residents towards constructed wetlands. The study was conducted with the help of a questionnaire which was sent to 1000 Vihti residents. The valuation was conducted with the choice experiment method. There were four different attributes in total: 1. recreational possibilities, 2. number of plant and animal species, 3. the state of the Lake Enäjärvi and 4. informing. The used payment vehicle was annual stormwater fee for a household. Questionnaires of this study where sent in three different phases during the summer 2016. The survey received 277 responses during the five-week time it was open. The response rate of the survey was 27,7%. Responses were gathered by mail and on a web-based survey. Valuation results of this study were analyzed with the Multinomial Logit-model. All together two different models were made. The first one had only the valuated attributes in it whereas the second model also included different background information. For the valuation we could draw a conclusion that the respondents where willing to pay fairly big amounts for certain attributes. They were the most willing to pay for the improvement in the state of Lake Enäjärvi. The second most valuated attribute was the recreational possibilities followed by informing and the increase in the amount of plant and animal species. Of the valuated attributes, all of them, exept informing, were statistically significant at all times in the level of 1–5%. Background information had an impact on the willingness to pay to some extent. For example a person who had previously visited a constructed wetland was more eager than an average respondent to pay for developing constructed wetlands. The WTP also increased if the respondeds household had higher income and he/she was planning to live more than five years in Vihti. WTP decreased if the respondent was male and he/she lived in a detached house. Results from the questions measuring respondents’ attitudes, perceptions and beliefs reveals that most of the respondents’ attitudes towards constructed wetlands are positive and they believe that constructed wetlands create benefits to the surrounding areas. The most beneficial aspect which urban wetlands were felt to provide was the benefit of views of an augmented landscape. 72% of the respondents indicated willingness to participate in water environment conservation activities during the next five years. Finally, most of the respondents felt that the results of the Urban Oases project and it presence in Vihti were positive.
